DCF方案為晚期胃及胃食管結(jié)合部腺癌一線治療的可選方案。適用于ECOG 0～1分，體能狀態(tài)良好的晚期胃癌患者。該研究證據(jù)源于V325研究，該研究為一項(xiàng)由美國(guó)、歐洲、亞洲、拉丁美洲等34個(gè)研究中心開(kāi)展的國(guó)際多中心Ⅲ期隨機(jī)對(duì)照試驗(yàn)，旨在確立以多西他賽為基礎(chǔ)的聯(lián)合化療在晚期胃癌及胃食管結(jié)合部位腺癌中的作用與地位。結(jié)果發(fā)現(xiàn)：DCF方案延長(zhǎng)了患者無(wú)疾病進(jìn)展期及生存期，提高了生活質(zhì)量。但在獲得高效的同時(shí)，該方案也帶來(lái)了更高的毒性：DCF方案的臨床實(shí)際應(yīng)用受到限制。

而我國(guó)由15家胃癌治療中心開(kāi)展的mDCF方案治療晚期胃癌中國(guó)注冊(cè)臨床研究是針對(duì)中國(guó)人群的V325研究的橋接試驗(yàn)，下調(diào)劑量的mDCF方案的治療組在療效近似的同時(shí)不良事件均較V325研究中的發(fā)生率下降。該研究根據(jù)中國(guó)患者特征調(diào)整了劑量，符合中國(guó)患者特點(diǎn)。
